Official Full Name: TNF receptor associated factor 2
Also known as: TRAP; TRAP3; RNF117; MGC:45012
The protein encoded by this gene is a member of the TNF receptor-associated factor (TRAF) protein family. TRAF proteins associate with members of the TNF receptor superfamily and mediate their signal transduction. This protein directly interacts with the TNF receptor and forms a heterodimeric complex with TRAF1. This protein is required for TNF-α-mediated activation of MAPK8/JNK and NF-kappaB. The protein complex formed by this protein and TRAF1 interacts with inhibitors of apoptosis proteins (IAPs) and acts as a mediator of TNF receptor anti-apoptotic signaling. Interaction of this protein with TRADD, a TNF receptor-associated apoptosis signaling transducer, ensures the recruitment of IAPs, thereby directly inhibiting caspase activation. BIRC2/c-IAP1 is an apoptosis inhibitor with ubiquitin ligase activity, which can ubiquitinate and induce the degradation of this protein, thereby enhancing TNF-induced apoptosis. Multiple alternatively spliced transcript variants of this gene have been identified, but the biological validity of only one transcript has been determined.
