Vesicle-mediated protein sorting plays an important role in the segregation of intracellular molecules into distinct organelles. Genetic studies in yeast have identified more than 40 vacuolar protein sorting (VPS) genes involved in vesicle transport into the vacuole. This gene is a member of the Sec1 domain family and shares high sequence similarity with mouse, rat and yeast Vps45. The gene's exact function is unknown, but its high expression in peripheral blood mononuclear cells suggests a role in the transport of proteins, including mediators of inflammation. Multiple alternatively spliced transcript variants have been found for this gene.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
