1. Nucleic acid amplification and detection is one of the most valuable technologies in biological research today. Real-time detection of PCR products is achieved by adding a fluorescent chemical to the reaction that reports an increase in the amount of DNA as the proportion of fluorescent signal increases. Fluorescent chemistries used for this purpose include DNA-binding dyes and fluorescently labeled sequence-specific primers or probes.
2. Real-time fluorescent quantitative PCR technology can accurately and sensitively quantify the copy number of the starting template in a large dynamic range. Fluorescence amplification was monitored using a dedicated thermal cycler equipped with a fluorescence detection module. The MyiQ™ Single Color Real-Time PCR Detection System is built on the iCycler® thermal cycler to provide optimal PCR performance.
3. The MyiQ system uses a tungsten halogen lamp as the light source. The filter-based optical design allows the use of optimal wavelengths of light for excitation and emission, resulting in superior sensitivity and resolution between multiple fluorophores. The CCD detector captured images of all 96 wells simultaneously. This resulted in a comprehensive dataset illustrating the kinetic behavior of the data at each cycle.
