Endogenous proteins are produced and degraded in equilibrium, so their cellular levels are stable under stable environmental conditions. Crude cell extracts contain many endogenous enzymes, such as phosphatases and proteases, capable of degrading proteins in the extract. When proteins were extracted from cells and tissues in vitro, protein production was significantly inhibited and degradation was increased. The best way to increase the yield of intact protein is to add inhibitors of those enzymes that are known to exist. Protease inhibitor cocktails are used in cell lysates or tissue extracts to improve protein stability. The cocktail acts to inhibit proteases that degrade unphosphorylated or phosphorylated protein substrates.
