Karyologic analysis readily distinguishes dipteran cells from lepidopteran cells. Lepidopteran cells contain both macro- and microchromosomes, and the ploidy is quite variable. Isoenzyme analysis, employing the enzyme phosphoglucoisomerase (PGI) and isoelectric focusing (IEF), is used for both intragenic and intergenic identification of lepidopteran cell lines. Intragenic and intergenic differentiation of lepidopteran cell lines from genera is made using IEF and PGI. The technique of IEF and isoenzyme analysis is well suited for the identification of insect cell lines.
