Description

PROTEINDEX™ HiBond™ Ni-NTA Agarose 6FF has been optimized for automated purification applications. It has been widely used in the amplification and preparative purification of histidine-tagged proteins expressed in various hosts, such as E. coli, yeast, insect cells and mammalian cells.

The base matrix of PROTEINDEX™ HiBond™ Ni-NTA Agarose 6FF is highly cross-linked 6% agarose, which provides excellent chemical and mechanical stability for automated applications. The matrix carrier uses the chelating ligand nickel triacetic acid (NTA) to complex with nickel ions (Ni2+) to form a stable octahedral structure. The structure of the Ni-NTA complex is a compatible solution containing a certain concentration of reducing agent, denaturant, detergent and other additives.

The NTA ligand coordinates Ni2+ with four valences (tetradentate, coordination number 4), two of which can interact with the imidazole ring of histidine residues. The number of valences available seems to provide the best combination of binding efficiency and chemical stability for His-tag binding. The Ni-NTA ion chelating complex on our ProteIndex™ HiBond™ Ni-NTA resin has been carefully optimized to ensure the most efficient capture of His-tagged proteins for His-tagged purification.

The prepackaged cartridges have standard interfaces for use with the most commonly used chromatography systems, such as the ÄKTA system.
