The Peptide Sulfhydryl Immobilization Kit is designed for simple and efficient coupling of sulfhydryl-containing polypeptides to solid agarose supports. Thiol-coupled resin columns utilize iodoacetyl groups, which specifically react with free thiols to form covalent, permanent thioether linkages. Long spacer arms reduce steric hindrance, ensuring greater binding of protein and antibody during affinity purification.

Peptides must have free sulfhydryl groups in order to be immobilized on the resin. The provided Protein-S-S-Reductant™ Reductant effectively reduces disulfide bonds without interfering with iodoacetyl coupling reactions. The advantage of Protein-S-S-Reductant™ is that no removal is required prior to immobilization of the peptide.

The resulting column can be used to purify antibodies against specific peptides. Depending on the stability of the peptide, the column can be used multiple times without significant loss of activity.
