Description

The TALON PMAC Magnetic Phosphorylation Enrichment Kit combines the phosphate specificity of our TALON Phosphoprotein Enrichment Kit with the convenience of magnetic bead separation to provide a simple, rapid, metal affinity-based method for Isolate microgram quantities of phosphorylated proteins from mammalian cells and tissues. The magnetic particles in the beads facilitate quick and easy purification of trace amounts of phosphoproteins when placed on a magnetic separator.

Isolation of cytoplasmic and membrane-bound proteins

Each kit provides a complete set of buffers, as well as our phosphate magnetic beads for population-specific enrichment of all types of phosphoproteins, including cytoplasmic and membrane bound, containing phosphorylated amino acid side chains, including serine, Tyrosine or threonine.
The extraction/loading buffer contains non-ionic detergents for the extraction of cellular proteins. Wash and elution buffers are detergent-free. If desired, phosphoproteins can be eluted in pH 9.0 buffer, which is more suitable for downstream applications.

Microscale purification in small elution volumes

Phosphomagnetic beads are supplied as a 5% suspension with a binding capacity of 400µg α-casein per mL of suspension. Different numbers of beads can be used, depending on the initial sample concentration. Phosphoproteins can be eluted in small volumes (50-200 μl) to produce concentrated samples.

Highly selective enrichment of phosphoproteins

Of all cellular proteins, only a small fraction is phosphorylated, so it is often necessary to enrich for this fraction before starting the analysis. The TALON PMAC Magnetic Phosphorus Enrichment Kit can be used with any mammalian cell type or tissue sample. The yield of phosphorylated protein varies between cell lines. The concentration process is efficient.
