Chitinase 3-like protein 1 (CHI3L1) is secreted by many cell types, including macrophages, chondrocytes, and certain tumor cells. The exact physiological function of CHI3L1 is unknown, but it is involved in developmental and inflammatory diseases such as asthma and tumors. CHI3L1 is a Th2-promoting cytokine expressed at high levels in the tumor microenvironment and serum of cancer patients. Elevated CHI3L1 expression is strongly associated with the developmental stage and outcome of multiple types of tumors, suggesting that CHI3L1 may serve as a biomarker of disease severity. Treatment with a neutralizing antibody to CHI3L1 was effective in an animal model of glioblastoma multiforme. CHI3L1 can promote angiogenesis through VEGF-dependent and independent pathways. CHI3L1 also enhanced tumor survival under gamma irradiation.
