Analysis of CXCL3/GRO-γ by ELISA (CAT#: STEM-MB-1562-LGZ)

Introduction

CXCL1, CXCL2, and CXCL3, known as GRO alpha, GRO beta (MIP-2 alpha), and GRO gamma (MIP-2 beta), respectively, are members of the CXC subfamily of chemokines. The mature CXCL1/2/3 protein is highly affinitively bound to IL-8 receptor type B and is an effective neutrophil attractor and activator. Chemokines mainly act on leukocytes and regulate their transport. In general, members of the chemokine α family preferentially chemotactic and activate neutrophils. Recombinant Human Growth Regulatory Oncogene γ(GRO-γ) is a 7.9kDa protein composed of 73 amino acids, including the "ELR" modle shared by the CXC chemokine family that binds to CXCR1 or CXCR2. The amino acid sequence homology of mature human CXCL3 (aa 35-107) and mouse CXCL3 and rat CXCl3 was 70% and 74%, respectively. CXCL3 plays a key role in monocyte migration and adhesion through its interaction with the CXCR2 receptor.




Principle

Enzyme-linked immunosorbent assay (ELISA) is an enzyme-labeled solid phase immunoassay technique. Its basic principle is to bind the antigen (or antibody) to the solid phase carrier, and the antigen (or antibody) and a certain enzyme link to enzyme labeled antigen (or antibody). During detection, the sample to be tested and the enzymic antigen (or antibody) react with the antigen (or antibody) on the solid phase carrier according to certain procedures, and then remove the unreacted part by washing method. After adding the substrate, the substrate is catalyzed by the enzyme on the solid phase carrier to produce colored substances. Through qualitative or quantitative detection of the amount of colored products, the content of the substance to be measured in the sample can be determined.

Procedure

1. Add standards or samples to each well and incubate.
2. Pour off the liquid in the well, biotinylated antibody working solution and incubate.
3. Add enzyme conjugate working solution and incubate.
4. Add substrate TMB and incubate.
5. Add stop solution and measure OD value.
6. Calculation of results.
