Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and assist in protein synthesis within mitochondria. Mitochondrial ribosomes consist of a small 28S subunit and a large 39S subunit. They are estimated to be composed of 75% protein and rRNA compared to prokaryotic ribosomes, whose proportions are reversed. Another difference between mammalian ribosomes and prokaryotic ribosomes is that the latter contain 5S rRNA. The proteins that make up the mitochondrial body vary widely in sequence and sometimes in biochemical properties across species, making sequence homology difficult to identify. This gene encodes the 28S subunit protein, which is also involved in the apoptotic pathway triggered by tumor necrosis factor-alpha, Fas ligand, and interferon-gamma. This protein may bind ATP/GTP and may be a functional partner of mitochondrial protein S27. Multiple alternatively spliced transcript variants encoding different isoforms of this gene have been found. Pseudogenes corresponding to this gene are located on chromosomes 1q and 2q.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
