Analysis of DICER1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2493-LGZ)

Introduction

Official Full Name: dicer 1, ribonuclease III
Also known as: DCR1; GLOW; MNG1; aviD; Dicer; HERNA; RMSE2; Dicer1e; K12H4.8-LIKE
This gene encodes a protein with an RNA helicase motif containing a DEXH box at its amino terminus and an RNA motif at its carboxy terminus. The encoded protein functions as a ribonuclease and is required for the RNA interference and small temporal RNA (stRNA) pathways to generate active small RNA components that repress gene expression. The protein is also a powerful antiviral drug, active against RNA viruses including Zika and SARS-CoV-2. Alternative splicing results in multiple transcript variants.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
