Analysis of F5 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0163-LGZ)

Introduction

Also known as: FVL; PCCF; THPH2; RPRGL1<br />This gene encodes an important cofactor of the coagulation cascade. The factor circulates in the plasma and is converted to the active form by the thrombin-releasing activating peptide during coagulation. This creates heavy and light chains linked together by calcium ions. Activin is a cofactor involved in the activation of coagulation factor X to activate prothrombin to thrombin. Defects in this gene result in autosomal recessive hemorrhagic or autosomal dominant thrombosis, ie, activated protein C resistance.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
