Introduction

Official Full Name: glutamate-ammonia ligase<br />Also known as: GS; GLNS; PIG43; PIG59<br />The protein encoded by this gene belongs to the glutamine synthetase family. It catalyzes the synthesis of glutamine from glutamate and ammonia in an ATP-dependent reaction. This protein plays a role in ammonia and glutamate detoxification, acid-base balance, cell signaling and cell proliferation. Glutamine is an abundant amino acid that is essential for the biosynthesis of several amino acids, pyrimidines and purines. Mutations in this gene are associated with congenital glutamine deficiency, and overexpression of this gene was observed in some primary liver cancer samples. Six pseudogenes of this gene were found on chromosomes 2, 5, 9, 11 and 12. Alternative splicing results in multiple transcript variants.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
