Analysis of HNF1B Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2488-LGZ)

Introduction

Official Full Name: HNF1 homeobox B
Also known as: T2D; FJHN; HNF2; LFB3; RCAD; TCF2; HPC11; LF-B3; MODY5; TCF-2; VHNF1; ADTKD3; HNF-1B; HNF1beta; HNF-1-beta
This gene encodes a member of the homeodomain-containing transcription factor superfamily. The protein binds to DNA as a homodimer or heterodimer, and binds to the related protein hepatocyte nuclear factor 1-α. This gene has been shown to play a role in nephron development and regulates the development of the embryonic pancreas. Mutations in this gene lead to kidney cysts, diabetic syndrome, and non-insulin-dependent diabetes, and the expression of this gene is altered in some types of cancer. Multiple transcript variants of this gene have been found encoding different isoforms.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
