The insulin-like growth factor (IGF) binding protein superfamily includes six high-affinity IGf-binding proteins (IGFBP) and at least four low-affinity binding proteins known as igFBP-associated proteins (IGFBP-RP). All IGFBP superfamily members are cysteine-rich proteins with conserved cysteine residues. IGFBPs help extend the half-life of IGFs in all tissues, including the prostate. In humans, insulin-like growth factor binding protein 6(IGFBP-6) is a protein encoded by the IGFBP6 gene. IGFBP-6 binds to IGF-II with a higher affinity than IGF-I, and it is a specific inhibitor of IGF-II. IGFBP-6 has been reported to play a role in cell proliferation, survival, differentiation, and migration independent of IGF. IGFBP-6 is unique among IGFBPs due to its clear IGF-II binding preference.
