Analysis of RAD51B Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2495-LGZ)

Introduction

Official Full Name: RAD51 paralog B
Also known as: REC2; R51H2; RAD51L1
The protein encoded by this gene is a member of the RAD51 protein family. RAD51 family members are evolutionarily conserved proteins required for DNA repair by homologous recombination. This protein has been shown to form stable heterodimers with family member RAD51C and further interact with other family members such as RAD51, XRCC2 and XRCC3. Overexpression of this gene leads to cell cycle G1 delay and apoptosis, suggesting a role for this protein in sensing DNA damage. Rearrangements between this locus and high mobility group AT-hook 2 have been observed in uterine leiomyomata.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
