Introduction

Official Full Name: ret proto-oncogene<br />Also known as: PTC; MTC1; HSCR1; MEN2A; MEN2B; CDHF12; CDHR16; RET-ELE1<br />This gene encodes a transmembrane receptor, a member of the tyrosine protein kinase family. Ligands such as GDNF (glial cell line-derived neurotrophic factor) and other related proteins bind to encoded receptors, stimulate receptor dimerization, activate downstream signaling pathways, and play a role in cell differentiation, growth, migration, and survival. The encoded receptors play important roles in the development of the nervous system and in the development of neural crest-derived organs and tissues. This proto-oncogene can undergo oncogenic activation through cytogenetic rearrangements and activating point mutations. Mutations in this gene are associated with Hirschsprung disease and central hypoventilation syndrome, and have been found in patients with renal agenesis.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
