The Anti-Human CD137 mAb manufactured by Seafrom is suitable for promoting the activation and expansion of T cells in cell therapy. 4-1BB, also known as CD137, is an inducible cell surface receptor and belongs to the tumor necrosis factor receptor (TNFR) superfamily. It is encoded by the tumor necrosis factor receptor superfamily member 9 (TNFRSF9) gene. CD137 is widely expressed on the surface of activated T cells, NK cells, and other human immune cells. Its ligand is CD137L, which also belongs to the TNF superfamily. CD137L is an inducible co-stimulatory receptor expressed on activated CD4+ and CD8+ T cells, NKT cells, NK cells, dendritic cells, macrophages, eosinophils, neutrophils, and mast cells, especially on CD8+ T cells. CD137/CD137L regulates T cell-mediated immune responses by transmitting activation, proliferation, or apoptosis signals between immune cells. Activation of CD137 can stimulate NK cells and T cells to proliferate and produce anti-tumor activity, leading to the generation of long-lasting memory responses. Using an agonistic 4-1BB monoclonal antibody to activate 4-1BB, it recruits TNFR-associated factors TRAF1 and TRAF2 to form a heterotrimer through pathways like c-Jun N-terminal kinase (JNK) and extracellular signal-regulated kinase (ERK). It enhances signal transduction and regulates 4-1BB signaling through the master transcription factor NF-κB, stimulating T cells and antigen-presenting cells to proliferate and secrete cytokines, thereby enhancing the body's anti-tumor immune response.
