DNA shuffling to generate mutant genes (CAT#: STEM-MB-0083-WXH)
Introduction
DNA shuffling, also known as molecular breeding, is an in vitro random recombination method to generate mutant genes for directed evolution and to enable a rapid increase in DNA library size.
This technique depends on the homologous gene fragments can be randomly recombined by DNaseI or restriction enzymes. Up to now, DNA shuffling is a powerful tool used to increase mutagenesis librari size.
Applications
Applied to protein and small molecule pharmaceuticals, bioremediation, vaccines, gene therapy, and evolved viruses.
Procedure
1. Acquiring DNA fragments for shuffling 2. Reassembly of DNaseI fragments 3. Aamplificaiton of full-length sequences
