Guaiacol Peroxidase Analysis in Plant Tissues

All kinds of abiotic and biotic stresses significantly limit plant productivity by disturbing cellular biochemistry and physiology via the overproduction of activated oxygen molecule-reactive oxygen species (ROS). Uncontrolled accumulation of ROS results in oxidation of DNA and RNA, protein denaturation and decreased enzyme activity as well as lipid peroxidation and carbohydrate oxidation.

Guaiacol peroxidase (GPOD) is heme-containing protein that catalyzes oxidation of aromatic electron donors such as guaiacol and pyragallol by oxygen of H2O2. Antioxidant function of guaiacol peroxidase consists in detoxification of hydrogen peroxide as one of ROS. The enzyme is localized in the vacuoles, the system of vesicular transport, and the cell wall. GPOD is an important component in the plant defense system against abiotic stresses. Many researches showed that the activity of GPOD is induced by various pollutants came from environment, such as heavy metals, herbicides, ozone, and polycyclic aromatic hydrocarbon.
