CD16 is a low-affinity Fc receptor, identified as Fc receptors FcγRIIIa (CD16a) and FcγRIIIb (CD16b). These receptors specifically bind to the Fc portion of IgG antibodies. CD16 is encoded by two highly homologous genes in a cell type-specific manner. It is present on the surface of natural killer cells, neutrophil polymorphonuclear leukocytes, monocytes, and macrophages. CD16a, also known as Low-affinity immunoglobulin gamma Fc region receptor III-A (FcγRIIIa), is an intermediate-affinity polypeptide-anchored transmembrane glycoprotein expressed on natural killer cells, macrophages, a subpopulation of T-cells, immature thymocytes, and placental trophoblasts. CD16b is a low-affinity, GPI-linked receptor expressed by neutrophils and eosinophils.
CD16a is involved in various immune functions, including phagocytosis, secretion of enzymes and inflammatory mediators, antibody-dependent cytotoxicity, and clearance of immune complexes. Aberrant expression or mutations in CD16a are implicated in susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.
The lyophilized form of CD16 can be stored at 4°C for 24 months, and the reconstituted liquid can be stored at -20°C for 6-12 months, avoiding repeated freeze-thaw cycles.
