Single-domain antibody (SdAb) contains only a single antibody variable region (VH/VL). Like conventional antibodies, it is also capable of selectively binding to specific antigens. The molecular weight of a single domain antibody is only 12-15 kDa, which is much smaller than that of an ordinary antibody (150-160 kDa) consisting of two heavy chains and two light chains.The first single domain antibodies were discovered from camelids. There is an antibody in the serum of camels that naturally lacks light chain and heavy chain constant region 1 (constant region 1, CH1), called heavy chain antibody (heavy chain antibody, HcAb). Single-domain antibodies are being researched for multiple pharmaceutical applications, and have potential for use in the treatment of acute coronary syndrome, cancer, Alzheimer's disease, and Covid-19.
