Isothermal titration calorimetry (ITC) is currently a valuable method for determining the thermodynamic constants, association constants, and stoichiometry of molecular interactions in aqueous solutions. This technique has been widely used by biochemists to study the interactions of proteins with other proteins, small molecules, metal ions, lipids, nucleic acids, and carbohydrates; and the interactions of nucleic acids with small molecules. The drug discovery industry already utilizes this method to measure the interaction of proteins (or nucleic acids) with drug candidates. ITCs have been used to screen drug candidates, guide the design of potential drugs, and validate modeling used in structure-based drug design.
