Analysis of CDC73 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0171-LGZ)

Introduction

Also known as: HYX; FIHP; HPTJT; HRPT1; HRPT2; C1orf28<br />This gene encodes a tumor suppressor and is involved in transcriptional and post-transcriptional control pathways. This protein is a component of the PAF protein complex, associated with the RNA polymerase II subunit POLR2A and the histone methyltransferase complex. This protein appears to facilitate the association of 3' mRNA processing factors with actively transcribed chromatin. Mutations in this gene have been linked to hyperparathyroidism-jaw tumor syndrome, familial isolated hyperparathyroidism, and parathyroid carcinoma.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Hyperparathyroidism 1, Hyperparathyroidism 2 with jaw tumors, Parathyroid carcinoma

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
