Analysis of CDKN2C Gene (Mutation) in Multiple Myeloma by RT-qPCR (CAT#: STEM-MT-0108-LGZ)

Introduction

Also known as: p18; INK4C; p18-INK4C<br />The protein encoded by this gene is a member of the INK4 family of cyclin-dependent kinase inhibitors. This protein has been shown to interact with CDK4 or CDK6 and prevent the activation of CDK kinases, thus functioning as a cell growth regulator controlling cell cycle G1 progression. Ectopic expression of this gene inhibits human cell growth in a manner that appears to be correlated with the presence of wild-type RB1 function. Studies in knockout mice have shown a role for this gene in regulating spermatogenesis as well as suppressing tumorigenesis. Two alternatively spliced transcript variants of this gene, encoding the same protein, have been reported.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
