Muscarinic cholinergic receptors belong to a larger family of G protein-coupled receptors. The functional diversity of these receptors is determined by acetylcholine binding and includes cellular responses such as adenylyl cyclase inhibition, phosphocarnosine denaturation, and potassium channel mediation. Muscarinic receptors influence many of the actions of acetylcholine in the central and peripheral nervous systems. Muscarinic cholinergic receptor 3 controls smooth muscle contraction and its stimulation causes secretion from glandular tissue. Different promoter usage and different splicing lead to multiple transcript variants with different tissue specificities.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
