Analysis of FGL1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1918-LGZ)

Introduction

Official Full Name: fibrinogen like 1
Also known as: HPS; HFREP1; HP-041; LFIRE1; LFIRE-1
Fibrinogen-like 1 is a member of the fibrinogen family. This protein is homologous to the carboxy-terminus of the β- and γ-subunits of fibrinogen containing four conserved cysteines of fibrinogen and fibrinogen-related proteins. However, this protein lacks platelet-binding sites, cross-linking regions, and thrombin-sensitive sites that are required for fibrin clot formation. This protein may play a role in the development of HCC. Four alternatively spliced transcript variants encoding the same protein are present in this gene.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
