Analysis of RAB13 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0601-LGZ)

Introduction

Official Full Name: RAB13, member RAS oncogene family
Also known as: GIG4
This gene is a member of the Rab family of small G proteins and plays a role in regulating membrane trafficking between the trans-Golgi network (TGN) and circulating endosomes (RE). The encoded protein is involved in the assembly of tight junctions, which are components of the apical junctional complex (AJC) in epithelial cells. The AJC plays a role in forming a barrier between the luminal contents and the underlying tissue. Other functions associated with this protein include endocytic recycling of occludins, regulation of epithelial cell scattering, neuronal regeneration, and regulation of neurite outgrowth. Alternately spliced transcript variants of this gene have been observed. A pseudogene related to this gene is located on chromosome 12.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
