Principle

Glutathione S-transferase (GST) has the ability to catalyze the binding of reduced glutathione (GSH) to dinitrobenzene (CDNB). The activity of the enzyme is indicated by measuring the binding rate of substrate GSH to dinitrobenzene in unit time. Yellow thio-nitrobenzoic acid anion (TNB) was generated by the reaction of the remaining GSH with dithio-di-dinitrobenzoic acid (DTNB). The reduction of GSH was calculated by measuring the concentration of the anion. Thus, the enzyme activity of glutathione S- transferase (GST) was calculated.

Applications

It is used to detect the enzyme activity of GST in serum (plasma) and animal tissues.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
