Analysis of IGF2 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2267-LGZ)

Introduction

Official Full Name: insulin like growth factor 2
Also known as: GRDF; SRS3; IGF-II; PP9974; C11orf43
This gene encodes a member of the insulin polypeptide growth factor family, involved in development and growth. It is an imprinted gene expressed exclusively from the paternal allele, and epigenetic alterations at this locus have been associated with Wilms tumor, Beckwith-Wiedemann syndrome, rhabdomyosarcoma, and silverrussell syndrome. There is a readable INS-igf2 gene whose 5' region overlaps with the INS gene and its 3' region overlaps with the INS gene. Alternative splice transcript variants of this gene have been found to encode distinct isoforms.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
