Analysis of JAK1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0113-LGZ)

Introduction

Also known as: JTK3; AIIDE; JAK1A; JAK1B<br />This gene encodes a membrane protein that is a member of a class of protein tyrosine kinases (PTKs) characterized by the presence of a second phosphotransferase-associated domain at the N-terminus of the PTK domain. The encoded kinase phosphorylates STAT proteins (signal transducers and activators of transcription) and plays a key role in interferon-α/β, interferon-γ and cytokine signaling. This gene plays a critical role in the expression of genes mediating inflammation, epithelial remodeling, and metastatic cancer progression. This gene is a key component of the interleukin-6 (IL-6)/JAK1/STAT3 immune and inflammatory response and is a therapeutic target to mitigate cytokine storm. The kinase activity of this gene is directly repressed by the suppressor of cytokine signaling 1 (SOCS1) protein. Alternative splicing results in multiple transcript variants.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
