Analysis of MLH3 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2494-LGZ)

Introduction

Official Full Name: mutL homolog 3<br />Also known as: HNPCC7<br />This gene is a member of the multiple homology (MLH) family of DNA mismatch repair (MMR) genes. The MLH gene is involved in maintaining genome integrity during DNA replication and meiotic recombination. The protein encoded by this gene functions as a heterodimer with other family members. Somatic mutations of this gene frequently occur in tumors exhibiting microsatellite instability, and germline mutations are associated with hereditary nonpolyposis type 7 colorectal cancer (HNPCC7). Several alternatively spliced transcript variants have been identified, but the full-length nature of only two transcript variants has been determined.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
