Analysis of PPM1B Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1759-LGZ)

Introduction

Official Full Name: protein phosphatase, Mg2+/Mn2+ dependent 1B
Also known as: PP2CB; PP2CBETA; PP2C-beta; PPC2BETAX; PP2C-beta-X
The protein encoded by this gene is a member of the PP2C family of serine/threonine protein phosphatases. Members of the PP2C family are known to be negative regulators of cellular stress response pathways. This phosphatase has been shown to dephosphorylate cyclin-dependent kinases (CDKs) and thus may be involved in cell cycle control. Overexpression of this phosphatase has been reported to lead to cell growth arrest or cell death. Alternative splicing results in multiple transcript variants encoding different isoforms. Other transcript variants have been described but do not currently represent full-length sequences.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
