Analysis of PTPRC Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0606-LGZ)

Introduction

Official Full Name: protein tyrosine phosphatase receptor type C
Also known as: LCA; LY5; B220; CD45; L-CA; T200; CD45R; GP180; IMD105
The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes, including cell growth, differentiation, mitosis, and oncogenic transformation. This PTP contains an extracellular domain, a transmembrane segment, and two tandem intracytoplasmic catalytic domains and is therefore classified as a receptor-type PTP. This PTP has been shown to be an important regulator of T-cell and B-cell antigen receptor signaling. It acts through direct interaction with components of the antigen receptor complex or through activation of various Src family kinases required for antigen receptor signaling. This PTP also inhibits JAK kinases and thus acts as a regulator of cytokine receptor signaling. Alternately spliced transcript variants of this gene encoding different isoforms have been reported.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
