Analysis of SDHC Gene (Mutation) in Hereditary Pheochromocytoma-Paraganglioma by RT-qPCR (CAT#: STEM-MT-0157-LGZ)

Introduction

Also known as: CYBL; PGL3; QPS1; SDH3; CYB560<br />This gene encodes one of four nuclear-encoded subunits that make up succinate dehydrogenase, also known as mitochondrial complex II, a key enzyme complex of the mitochondrial tricarboxylic acid cycle and the aerobic respiratory chain. The encoded protein is one of two integral membrane proteins that anchor the other subunits of the complex forming the catalytic core to the inner mitochondrial membrane. This gene has several related pseudogenes on different chromosomes. Mutations in this gene are associated with paragangliomas. Alternative spliced transcript variants have been described.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
