Analysis of SLC34A3 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2025-LGZ)

Introduction

Official Full Name: solute carrier family 34 member 3<br />Also known as: HHRH; NPT2C; NPTIIc<br />This gene encodes a member of the SLC34A transporter family and is mainly expressed in the kidney. It is involved in the transport of phosphate into cells by sodium cotransport through the renal brush border membrane and helps maintain the concentration of inorganic phosphate in the kidney. Mutations in this gene are associated with hereditary hypophosphatemic rickets with hypercalciuria. Different spliced transcript variants were found in the 5'UTR of this gene.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
