Analysis of SP100 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1540-LGZ)

Introduction

Official Full Name: SP100 nuclear antigen<br />Also known as: lysp100b<br />This gene encodes subnuclear organelles and major components of the PML (promyelocytic leukemia)-SP100 nucleosome. PML and SP100 are covalently modified by SUMO-1 modifiers, which are thought to be critical for nucleosomal interactions. The encoded protein binds heterochromatin proteins and is thought to play a role in tumorigenesis, immunity, and gene regulation. Alternative splice variants of this gene have been identified; one of them encodes a high mobility protein cluster.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
