Analysis of SPEN Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1423-LGZ)

Introduction

Official Full Name: spen family transcriptional repressor
Also known as: MINT; SHARP; RATARS; RBM15C; HIAA0929
This gene encodes a hormone-induced transcriptional repressor. Transcriptional repression of this gene product can be achieved through interactions with other repressors, through recruitment of proteins involved in histone deacetylation, or through sequestration of transcriptional activators. The product of this gene contains a carboxy-terminal domain that allows binding to other corepressor proteins. This domain also allows interaction with members of the NuRD complex, a nucleosome remodeling protein complex with sirtuin activity. Furthermore, this suppressor contains several RNA recognition motifs that confer binding to steroid receptor RNA coactivators; this binding can modulate the activity of both liganded and unliganded steroid receptors.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
