Analysis of UBAP1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1954-LGZ)

Introduction

Official Full Name: ubiquitin associated protein 1<br />Also known as: UAP; UBAP; NAG20; SPG80; UBAP-1<br />This gene is a member of the UBA domain family, whose members include proteins associated with the ubiquitin and ubiquitination pathways. The ubiquitin-associated domain is considered a non-covalent ubiquitin-binding domain consisting of a tight triple-helical bundle. This particular protein originates from a locus in the refinement region of chromosome 9 that occurs in nasopharyngeal carcinoma (NPC) where loss of heterozygosity occurs. Given its cytogenetic location, this UBA domain family member is being investigated as a putative mutational target in nasopharyngeal carcinoma. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
