A normalized cDNA library is a cDNA library constructed by reducing the number of copies of redundant transcripts and increasing the number of copies of low-abundance transcripts, in which all expressed genes of a particular tissue or cell are included and the number of copies of cDNAs corresponding to expressed genes in the cDNA library is equal or close. Normalized cDNA libraries are an effective measure to overcome the barriers to library screening and analysis caused by the great differences in gene transcript levels, and facilitate the study of gene expression and analysis.
Normalized cDNA libraries have the following four advantages.
1. Save the cost of large amount of experiments.
2. Increase the chance of cloning low-abundance mRNAs.
3. The expression level of most genes can be estimated and some tissue-specific genes can be identified.
4. Can be used for genetic mapping and large scale in situ hybridization and sequencing.
