Description

PROTEINDEX™ Anti-DYKDDDDK Sepharose 4 Fast Flow is an affinity chromatography medium for the purification of proteins containing the peptide sequence DYKDDDDK (n-asp-tir-lys-asp-asp-asp-lys-c). The ligand covalently immobilized on the matrix support is a recombinant mouse monoclonal IgG1 species that binds to the same epitope as Sigma's Anti-FLAG® M2 antibody and which is highly sensitive to the peptide DYKDDDDK in a calcium-independent manner specificity. Commonly referred to as the FLAG® epitope, the amino acid sequence DYKDDDDK is used as an affinity tag, fused at the N-terminus, C-terminus, or an internal position of the recombinant protein.

For protein purification, ProteIndex™ Anti-DYKDDDDK Sepharose 4FF Resin is added to the sample. After capturing dykddddk-tagged proteins, unbound and non-specifically bound proteins can be washed away. The bound target protein can be eluted with a competing peptide under mild conditions or with an acidic glycine-HCl buffer.

Since the binding between the epitope and the antibody can occur under natural conditions, any proteins or other macromolecules capable of interacting with the tagged fusion protein will also be captured during the binding step. Therefore, anti-dykddddk 4FF affinity resin can be used for co-immunoprecipitation to analyze protein complexes associated with fusion proteins of interest.

The base matrix of ProteIndex™ Anti-DYKDDDDK Agarose 4FF is highly cross-linked 4% agarose, which provides strong support for low background and high physical stability for chromatography procedures of all scales. Affinity resins are recommended for gravity/batch applications, spin cleanup columns, or cartridges for FPLC instruments.
