Description

PROTEINDEX™ rProtein A Sepharose 4 Fast Flow is an affinity chromatography medium designed for simple one-step purification of immunoglobulins, especially of the IgG isotype, and fusion proteins containing the Fc region.

Protein A is a bacterial cell wall protein isolated from Staphylococcus aureus that binds to the Fc region of most immunoglobulins through interaction with the heavy chain. Native protein A contains five high-affinity IgG-binding domains and many domains with unknown functions.

Recombinant protein A is a genetically engineered protein that contains only 5 high-affinity IgG binding domains. Non-essential domains were removed to minimize non-specific binding. Recombinant protein A is produced in E. coli and it binds immunoglobulins with essentially the same affinity as native protein A. Protein A can have a high affinity with the Fc region of IgG of various species such as human and rabbit.

Through the proprietary ProteIndex™ technology, the immobilization of recombinant protein A on 4% highly cross-linked agarose resin is very stable, resulting in high yields in laboratory preparations and scale-up applications.
