Study of Type IB Topoisomerases by Magnetic tweezers (MT) (CAT#: STEM-MB-1245-WXH)

Introduction

Type IB DNA topoisomerases are found in all eukarya, two families of eukaryotic viruses (poxviruses and mimivirus), and many genera of bacteria. They alter DNA topology by cleaving and resealing one strand of duplex DNA via a covalent DNA-(3-phosphotyrosyl)-enzyme intermediate.

Principle

Magnetic tweezers (MT) are scientific instruments for the manipulation and characterization of biomolecules or polymers. These apparatus exert forces and torques to individual molecules or groups of molecules. It can be used to measure the tensile strength or the force generated by molecules.

Applications

Study of mechanical properties of biological macromolecules like DNA or proteins in single-molecule experiments.
Study of the rheology of soft matter.
Study of force-regulated processes in living cells.

Procedure

1.Sample preparation
2.Force Calibration
3.Measurement
