Analysis of MYCNOS Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1125-LGZ)

Introduction

Official Full Name: MYCN opposite strand
Also known as: NCYM; NYCM; N-CYM; MYCN-AS1
The gene is transcribed antisensely with v-myc avian myeloma virus oncogene neuroblastoma homologous gene (MYCN). It is thought to encode a small, novel protein that stabilizes MYCN, prevents apoptosis, and promotes cell proliferation. Transcripts from this site can also directly serve as functional RNAs, recruiting transcriptional regulators to the promoter of MYCN and stimulating the transcription of this oncogene. Thus, the gene functions through both RNA and protein products.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Medulloblastoma, Glioblastoma, Pancreatic adenocarcinoma, Malignant neoplasm of body of uterus, Neuroblastoma

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
