Principle

According to different coenzyme specificities, MDH is divided into NAD-dependent MDH and NADP-dependent MDH. NAD-MDH usually exists in the cytoplasm and mitochondria of bacteria and eukaryotic cells. NAD-MDH malate dehydrogenase catalyzes the transformation of malic acid and NAD into oxaloacetate and NADH, and NADH makes WST-8 orange-yellow under the action of electron coupler. Malate dehydrogenase activity was calculated by measuring the change in absorbance at 450 nm.

Applications

It is suitable for detecting the activity of NAD- malate dehydrogenase (NAD-MDH) in serum (plasma), animal tissues and cells.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
