Introduction

Official Full Name: nectin cell adhesion molecule 4
Also known as: LNIR; PRR4; EDSS1; PVRL4; nectin-4
This gene encodes a member of the nectin family. The encoded protein contains two immunoglobulin-like (Ig-like) C2-type domains and one Ig-like V-type domain. It is involved in cell adhesion through trans-homophilic and -heterophilic interactions. It is a single generation type I membrane protein. The soluble form is produced by proteolytic cleavage of the cell surface by the metalloprotease ADAM17/TACE. The secreted form has been found both in breast tumor cell lines and in patients with breast tumors. Mutations in this gene are responsible for ectodermal dysplasia - and referred to as syndrome type 1, an autosomal recessive disorder. Alternatively spliced transcript variants have been identified, but the full-length nature of the variants has not been determined.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
