Analysis of PARK7 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1375-LGZ)

Introduction

Official Full Name: Parkinsonism associated deglycase
Also known as: DJ1; DJ-1; GATD2; HEL-S-67p
The product of this gene belongs to the peptidase C56 protein family. It is a positive regulator of androgen receptor-dependent transcription. It also acts as a redox-sensitive chaperone, as a sensor of oxidative stress, and it apparently protects neurons from oxidative stress and cell death. Defects in this gene are the cause of autosomal recessive early-onset Parkinson disease7. Two transcript variants of this gene have been identified encoding the same protein.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
