Introduction

Official Full Name: period circadian regulator 3<br />Also known as: GIG13; FASPS3<br />The gene, a member of the period gene family, is expressed in a circadian pattern in the suprachiasmatic nucleus, the major circadian pacemaker of the mammalian brain. Genes in this family encode circadian components of locomotor activity, metabolism, and behavior. The gene is upregulated by the CLOCK/ARNTL heterodimer, but then interacts with CLOCK/ARNTL through the PER/CRY heterodimer, inhibiting this upregulation in a feedback loop. Polymorphisms in this gene have been associated with sleep disorders. Multiple transcript variants of this gene have been found encoding different isoforms.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
