Analysis of RAD21 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1906-LGZ)

Introduction

Official Full Name: RAD21 cohesin complex component
Also known as: MGS; HR21; MCD1; NXP1; SCC1; CDLS4; hHR21; HRAD21
The protein encoded by this gene is highly similar to the gene product of Schizosaccharomyces pombe rad21, which is involved in the repair of DNA double-strand breaks and the cohesion of chromosomes during mitosis. This protein is a nuclear phosphorylated protein that is hyperphosphorylated during the M phase of the cell cycle. The highly regulated association of this protein with mitotic chromatin, particularly at the centromere region, suggests its role in sister chromatid cohesion in mitotic cells.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
